TIER A — At least one published randomized controlled trial in humans.
Randomized, placebo-controlled dose studies in healthy adults were published (2006); development was discontinued and it is not approved.
Tiers rate the strength of published evidence — not effectiveness, safety, or a recommendation.

Overview
A long-acting growth hormone releasing hormone analog with a drug-affinity complex that extends its half-life to several days. Provides sustained GH elevation with less frequent dosing.
Research status Phase II clinical trials completed; demonstrated sustained GH elevation in human subjects
Mechanism
CJC-1295 DAC is a synthetic GHRH analog with four amino acid substitutions for enzymatic stability plus a drug affinity complex (DAC) that binds albumin, extending the half-life to 6-8 days. It binds the GHRH receptor on pituitary somatotrophs, activating cAMP-PKA signaling to promote sustained GH secretion.
Effects reported in research
Reported outcomes, not promises. Some come from controlled human trials; others may be from smaller or preclinical studies — check the studies below.
- Sustained growth hormone elevation over days
- Improved body composition
- Enhanced recovery and deep sleep
- Increased IGF-1 levels
Safety & unknowns
Phase II trials showed generally well-tolerated profile. Common side effects include injection site reactions, water retention, and transient numbness/tingling. Sustained GH elevation may carry risks of prolonged IGF-1 increase. Not FDA-approved.
Unknowns. Trial safety data apply to the populations, formulations and conditions studied.
